Ac-Trp-Glu-His-Asp-pNA is a colorimetric substrate for Caspase 1. Caspase 1/Interleukin 1b converting enzyme (ICE) is a cysteine protease that preferentially cleaves substrates built on VAD (val-ala-asp) or WEAD (trp-glu-ala-asp) sequences.Ac-WEHD-pNA will also act as a substrate for caspase 4, caspase 5 and caspase 14. ICE converts inactive IL-1 beta precursor to a pro-inflammatory cytokine that mediates a wide range of immune and inflammatory response.ICE-related cysteine proteases are required for E1A-induced, p53-dependent apoptosis in baby rat kidney (BRK) cells. Adenovirus E1B 19K protein, which is a potent inhibitor of apoptosis, inhibits activation of these proteases in BRK cells.
Activity is quantified by the release of p-nitroaniline which is measured by absorbance at 405 nm.
