Dabcyl-LAQAVRSSSR-EDANS is a fluorescence energy transfer (FRET) peptide substrate for tumor necrosis factor (TNF)-α convertase (TACE) (Km ~19 µM). TACE, also known as ADAM 17, cleaves TNF-α from the membrane to activate it. TACE cleaves the Ala-Val bond and the substrate can be used to screen for inhibitors.
Activity is quantified by release of fluorescent EDANS which excites at 365 nm and emits at 490 nm.
